Simple fractionation for cleaner 2D gel maps

For a meaningful proteome analysis, the
proteome analysis must characterize and quantify all of the proteins in a
living system. In an animal cell system, there may be > 10,000 protein
species. Therefore, effective global analysis requires separation techniques
capable of resolving and detecting the majority, if not all, of the proteins
in cell systems. In whole cell lysate, there are proteins that exist in a
higher dynamic range of concentration; consequently, abundant proteins mask
identification of less abundant proteins. An effective proteome analysis
will require separation of abundant proteins, and concentration and
enrichment of less abundant proteins to bring the less abundant proteins
into the dynamic range of detection.
Fractionation has been proposed by many
researchers as a means of simplifying samples for better 2D resolution.
Fractionation allows preparation of pools of proteins containing fewer
protein species than found in the starting material.
Fractionation allows separation of highly expressed proteins from low
abundance proteins, bringing the low abundance proteins into the dynamic
range of detection.
Fractionation produces less crowded individual maps, simplifying analysis
and interpretation.
Fraction-FOCUSTM has been developed
to serially fractionate, concentrate and enrich proteins in a sample to
bring the less abundant proteins into the dynamic range of detection. The
method of fractionation is simple, robust, and contained within successive
fractionation tubes- without any waste or loss of the staring material.
Fraction-FOCUSTM exploits the
property of protein molecules to precipitate in response to the changes in
solvent composition (i.e., ionic strength, pH, temperature).
The fractionation involves just one step
for each fraction and there is no detectable loss of material. At the end of
the fractionation, cellular proteins are in one of many fractions. The
entire fractionation is carried out in micro scale.
