HOOKTM Activated Agaroses
For the
generation of peptide and protein affinity columns for the purification of
antibodies and for the discovery of important interacting proteins and cofactors.Genotech offers two activated agaroses for the coupling of
peptides and proteins to agarose through their primary amines or carboxy
and sulfhydryl residues. These kits generate stable bonds resulting in
columns that can be used several times.
HOOKTM Activated Agarose (Amine Reactive)
The amine reactive HOOKTM Activated Agarose is 6% agarose that has been
activated to generate reactive aldehyde groups. The aldehyde groups of the
agarose react spontaneously with primary amines, located at the N-terminus
of proteins or in lysine residues, to form intermediate Schiff Base
complexes. These, in turn, are selectively reduced by reductive amination
to form stable amine linkages between the agarose and the ligand.

HOOKTM Activated Agarose (Carboxy/Sulfhydryl
Reactive)
The carboxy/sulfhydryl reactive HOOKTM Activated Agarose is 6% agarose
beads that has been activated by the addition of an aminoethyl group.
Proteins and peptides are coupled to the agaroses amine group via the use
of common protein crosslinkers, such as EDC (Cat.# BC25) or sulfoSMCC
(Cat.# BC23). EDC will couple protein/peptide carboxyl groups to the
agarose, where as sulfoSMCC will react with the sulfhydryl residues of the
proteins/peptides.
Binding Capacity: 5mg protein/ml resin.
