An important tool in research is the generation of peptide and protein affinity columns for the purification of antibodies and for the discovery of important interacting proteins and cofactors. G-Biosciences offers a complete kit for the coupling of peptides and proteins to agarose through their sulfhydryl residues. These kits contain all the reagents to generate stable bonds resulting in columns that can be reused several times.
 
The sulfhydryl reactive HOOK Agarose Coupling kit utilizes our HOOK Activated Agarose (Carboxyl/sulfhydryl) resin that has a reactive aminoethyl group. The kit utilizes the supplied crosslinker sulfoSMCC to couple peptide sulfhydryl groups, found on cysteine side chains, to the aminoethyl on the agarose. SulfoSMCC is a heterobifunctional crosslinker that conjugates between sulfhydryl and primary amine residues. The N-hydroxysuccinimide (NHS) ester in sulfoSMCC reacts with primary amines to form covalent amide bonds and the maleimide group reacts with sulfhydryl groups to form stable thioether bonds. The resulting bonds are stable, allowing the columns to be reused.
 
In addition to all the coupling reagents, the HOOK Agarose Coupling Kit is supplied with a specific Optimizer Buffer that provides the optimal coupling conditions and can be used to equilibrate your starting protein sample.
 
This kit is suitable for the preparation of 5 stable columns containing 2ml resin.
 
If peptides do not contain a sulfhydryl group then we recommend our amine reactive HOOK Agarose Coupling Kit.


The kits allow for the rapid and simple production of affinity columns.
