HOOK™ AGAROSE COUPLING KIT (AMINE REACTIVE)

An important tool in research is the generation of peptide and protein affinity columns for the purification of antibodies and for the discovery of important interacting proteins and cofactors. G-Biosciences offers a complete kit for the coupling of peptides and proteins to agarose through their primary amines. These kits contain all the reagents to generate stable bonds resulting in columns that can be reused several times.
 
The amine reactive HOOK Agarose Coupling Kit is designed for the simple and efficient coupling of proteins and peptides to a solid support. The kit allows the generation of both a stable and reusable column for the affinity purification of antibodies, proteins and other protein interacting biomolecules.
 
This kit utilizes our HOOK Activated Agarose (Amine) that has reactive aldehyde groups. The aldehyde groups of the agarose react spontaneously with primary amines, located at the N-terminus of proteins or in lysine residues, to form intermediate Schiff Base complexes. These, in turn, are selectively reduced by the supplied reduction agent. Following quenching of the reaction the protein or peptide is linked to the agarose by stable amine linkage. The resin can now be aliquoted into the supplied columns for use in purification and interation studies.
 
In addition to all the coupling reagents, the HOOK Agarose Coupling Kit is supplied with a specific Optimizer Buffer that provides the optimal coupling conditions and can be used to equilibrate your starting protein sample.
 
This kit is suitable for the preparation of 5 stable columns containing 2ml resin.
