XIT™ Genomic DNA from Whole Blood

Protocols.io provides an interactive version of this protocol where you can discover and share optimizations with the research community.

The XIT Genomic DNA from Blood kit is designed for the isolation of genomic DNA from blood, bone marrow and buffy coat.

The kit uses four main steps (see figure) that are red blood cell lysis, rapid & efficient nuclear  lysis, complete protein precipitation, followed by DNA precipitation.  This yields high quality genomic DNA.

Three sizes of kit are available for processing a total of 12.5, 125 or 250ml of blood.  The purified DNA has a A260/A280 ratio between 1.7 and 1.9 and is up to 200kb in size.  The yield is approximately 10-15µg/ml of blood.

Features

  • Isolate high quality genomic DNA
  • Suitable for blood, bone marrow and buffy coat
  • Yield 10-15µg/ml blood
