Immobilized_Beta-cyclodextrin

Immobilized β-cyclodextrin is designed to allow refolding of recombinant proteins that were aggregated or formed inclusion bodies when expressed in the genetically engineered cells. The refolding of misfolded proteins is adapted from GroE chaperone refolding mechanism. The misfolded proteins or inclusion bodies are solubilized with detergent such as Triton X-100 in the first step to avoid protein aggregation. In the following step the detergent treated protein is passed through immobilized β-cyclodextrin agarose preequilibrated with refolding buffer. Cyclodextrin strips away the detergent allowing proper refolding of proteins.

  • Matrix: 6% cross-linked agarose
  • pH stability: 6-9
  • Chemical stability: Stable to all commonly used aqueous buffers and additives like detergents. Do not use azides.
  • Physical stability: Volume variations are negligible for changes in pH or ionic strength 
  • Storage temperature: 4°C
  • Supplied as 50% slurry in 20 % ethanol
