Immobilized Triton X-100

Immobilized_Triton_X-100

Immobilized Triton X-100 is designed to allow refolding of recombinant proteins that were aggregated or formed inclusion bodies when expressed in the genetically engineered cells. The refolding of misfolded proteins is adapted from GroE chaperone refolding mechanism. The misfolded proteins or inclusion bodies are solubilized with denaturants such as guanidine hydrochloride, 6M. The solubilized protein is captured by Triton X-100 when passed through immobilized Triton X-100 column. The elution of protein is carried out by increasing β-cyclodextrin concentration. β-cyclodextrin allows for proper refolding of the protein.

Features:

  • Active group: Triton X-100
  • Extent of labeling: 15-30 µmol /g of drained gel
  • Matrix: 6% cross-linked agarose
  • pH stability: 6-9
  • Chemical stability: Stable to all commonly used aqueous buffers and additives like detergents. Do not use azides.
  • Storage temperature: 4°C
  • Supplied as 50% slurry in 20 % ethanol

Application(s):

  • Immobilized Triton X-100 is use for refolding of recombinant proteins that were expressed in aggregated form or as inclusion bodies in genetically engineered cells  such as E. coli.
