Description
T7 Endonuclease I recognizes and cleaves mis-matched DNA. The cleavage takes place at the first, second or third phosphodiester bond that is 5´ to the mismatch. Mutations generated by CRISPR/ZFN/TALEN at desired target sites can be PCR-amplified, and the PCR products can be denatured and re-annealed to form mismatched DNA. T7 Endonuclease I can recognize and cleave mismatched DNA length more than 1bp. It is used to estimate the mutation frequency of genone edited cells.
