For mass spectrometry analysis complete digestion and recovery of peptides is needed. Residual silver ions used in traditional kits inhibit trypsin digestion. Glutaraldehyde, used as a sensitizer in many silver-staining methods, modifies lysine residues preventing complete digestion and recovery of the peptides.

FOCUS-FastSilverTM does not use protein modifier glutaraldehyde and the kit is provided with silver-ion removing and de-staining reagent SilverOUTTM which allows complete trypsin digestion and extraction of peptides needed for mass spectrometry.

FEATURES

- Full & Efficient Trypsin Digestion -
Silver has been found to inhibit trypsin digestion by binding at the active site of trypsin and chymotrypsin. FOCUS-FastSilverTM is provided with SilverOUTTM reagent for removal of silver ions from protein bands in polyacrylamide gels. This allows maximal trypsin digestion.
- Full and Efficient Recovery of Peptides - Glutaraldehyde modifies lysine residues and cross-link proteins, reducing peptide recovery. FOCUS-FastSilverTM does not use glutaraldehyde, used in traditional silver staining protocols. FOCUS-FastSilverTM allows enhanced trypsin digestion and efficient recovery of digested peptides.
