Using a Miniprep Kit for p-Amylase Plasmid Isolation (Lab 8G)

This kit is designed to teach 8 groups of four students the principles of plasmid DNA purification by minipreps.

The kit contains the reagents to allow students to:

• Collect p-Amylase-transformed bacteria from an overnight culture
• Resuspend bacteria, lyse and isolate the p-Amylase plasmid by removing proteins and genomic DNA
• Bind the plasmid DNA to a spin column and wash
• Elute the plasmid DNA from column
• Measure the amount of purified pAmylase plasmid DNA with a unique DNA assay
